The zero degree tilt view of the raw data acquired from an IVEM of a Caulobacter crescentus sample, strain Snc04. This image has been downsampled from the raw data image which can be accessed using the link provided to the Cell Centered Database.
Caulobacter crescentus (strain Snc04)were induced for 1 hour at 30 degrees, labeled for 2 hours at 30 degrees (in presence of 1.8 micromolar ReAsH-EDT2); post fixed with _0.01%_ Acrolein and 2.5% of Glut. in 0.01M Sodium cacodylate buffer; subjected to high pressure freezing and freeze substitution by using Epoxy resin as fixative: 20% of the complete Araldite/Epon formulation in acetone. Infiltration and embedding in 100% of resin complex. 0.5um sections were cut and collected on gold grids. Sections were post stained with 4% of Osmium followed by Sato lead and finally 10nm was applied on both sides of sections. Single tilt tomograms were obtained using a JEOL 4000, and spanned -52 - 66? at 2? intervals.
